Methotrexate ยานี้เป็นพิษต่อเซลล์ โดยต้านการทำงานของโฟลิกและรบกวนเมตาบอลิซึมของเซลล์ ค่าครึ่งชีวิต (half-life) ของยา 10-12 ชั่วโมง ยานี้ผ่านน้ำนมได้ ปริมาณที่ผ่านน้ำนมน้อย¹ แต่เนื่องจากยานี้สามารถสะสมในมารดาและทารกได้² ซึ่งการสะสมนี้อาจยาวนานถึง 1 ปี³ ดังนั้นจึงแนะนำให้หลีกเลี่ยงระหว่างการให้นมบุตร
